甜菜堿對(duì)脂肪組織釋放游離脂肪酸和抗氧化能力的影響

【摘要】 目的觀察甜菜堿對(duì)大鼠離體脂肪組織釋放游離脂肪酸(Free fatty acid, FFA)和抗氧化能力的影響,并探討其作用機(jī)制。方法將大鼠離體脂肪組織置于37℃ Krebs液中溫育,分別加入不同濃度的甜菜堿及各阻斷劑,觀察其對(duì)FFA和丙二醛(MDA)、總抗氧化能力(TAOC)的影響。結(jié)果甜菜堿可促進(jìn)脂肪組織釋放FFA(P=0.000),并存在劑量相關(guān)效應(yīng)(r=0.909,P=0.000);甜菜堿可降低脂肪組織MDA含量,提高TAOC;心得安、異搏定和SQ22,536(腺苷酸環(huán)化酶抑制劑)可不同程度的阻斷上述作用。結(jié)論甜菜堿可促進(jìn)離體脂肪組織釋放FFA,提高其抗氧化能力。此作用可能部分經(jīng)由脂肪細(xì)胞的腎上腺素能β受體、L型Ca2+通道和腺苷酸環(huán)化酶所介導(dǎo)。

Abstract:ObjectiveThe effect of betaine on free fatty acid(FFA) release and antioxidant capacity from isolated adipose tissue in rats and its mechanism were studied. MethodsIsolated rat tissues were placed into Krebs solution to incubate at 37℃ and were respectively added into the betaine of different concentration and blockers. the FFA values and malondialdehyde (MDA) and tatal antioxidant capacity(TAOC) content from isolated adipose tissue were observed. ResultsBetaine could promote the FFA release (P=0.001;r=0.909, P=0.000)dosedependently. It could reduce MDA and raise TAOC from isolated adipose tissue which can be blocked partly by propranolol, varapamil or SQ22,536(adenylyl cyclase inhibitor).ConclusionBetaine can enhance the catabolism of adipose tissue in vitro by increasing the release of FFA and improving its antioxidant capacity. The effect may be mediated partly by β3 adrenergic receptor, varapamilsensitive Ca2+ channel L and adenylyl cyclase(AC).

甜菜堿(betaine)存在于多種植物中,作為飼料添加劑,可抑制豬、雞的脂肪合成,提高瘦肉率[1]。還可調(diào)節(jié)滲透壓,降低脂肪組織的比率,防止脂肪肝,減輕乙醇所致的肝損傷,確保動(dòng)物正常生長(zhǎng)而無毒性[2]。甜菜堿作為一種甲基供體,有3個(gè)活性甲基,通過蛋氨酸循環(huán),參與蛋白質(zhì)、脂肪代謝[3]。本室前期的研究工作發(fā)現(xiàn)甜菜堿對(duì)高脂性肥胖大鼠體質(zhì)量有明顯的抑制作用,同時(shí)可降低高脂飲食大鼠血清MDA,提高TAOC。但甜菜堿對(duì)離體大鼠的脂肪組織是否有直接作用,國(guó)內(nèi)外尚未見文獻(xiàn)報(bào)道。本實(shí)驗(yàn)觀察甜菜堿對(duì)離體脂肪組織釋放FFA及抗氧化作用的影響,并初步探討其作用機(jī)制。
1 材料
1.1 藥品和溶液甜菜堿、小牛血清白蛋白(BSA)、腺苷酸環(huán)化酶抑制劑SQ 22,536由美國(guó)Sigma公司提供。心得安北京第二制藥廠生產(chǎn),異搏定蘭州制藥廠生產(chǎn),鹽酸腎上腺素天津金耀氨基酸有限公司提供。Krebs液成分如下:NaCl 7.05 g/ L,KCl 0.44 g/ L,NaH2PO4 0.187 g/ L,MgCl2 0.244 g/ L,NaHCO3 1.29 g/ L,CaCl2 0.277 g/ L,C6H12O6 1.14 g(未完,下一頁)
