芹菜甲素對(duì)D-半乳糖衰老模型小鼠的作用

（接上頁(yè)）續(xù)注射，小鼠可產(chǎn)生多項(xiàng)生化指標(biāo)的變化，如腦組織MDA、LF含量的升高；腦中SOD、血中GSH-PX、紅細(xì)胞中CAT活力的降低，這些變化和自然衰老的變化相一致，說(shuō)明D-半乳糖可引起細(xì)胞和機(jī)體的衰老[4]。SOD、GSH-PX、CAT活性間接反映機(jī)體清除氧自由基的能力；而MDA、LF水平高低間接反映細(xì)胞受自由基攻擊的嚴(yán)重程度。
本研究表明：芹菜甲素可以有效對(duì)抗D-半乳糖所致的小鼠亞急性衰老和學(xué)習(xí)記憶能力的降低，具有抗衰老作用。其抗衰老的機(jī)制可能是通過(guò)提高機(jī)體SOD、GSH-PX、CAT等抗氧化酶活力，從而對(duì)抗自由基對(duì)細(xì)胞的損傷，降低LPO、LF等代謝產(chǎn)物的產(chǎn)生而有效的對(duì)抗D-半乳糖所致的小鼠衰老體征的出現(xiàn)和學(xué)習(xí)記憶能力的下降。

Anti-aging action of Butyl-phthalide on D-galactose model mice

ABSTRACT OBJECTIVE By using the D-galactose-induced aging mice,the effects and mechanism of Butyl-phthalide on aging were investigated. METHODS Butyl-phthalide were given to mice by ig and D-galactose were injected into mice for six weeks. Then ,the ability of study of mice ,the content of LPO,LF,the body weight ,the index of thymus and spleen,the activities of superoxide dismutarase(SOD),catalase(CAT) and whole blood glutothine peroxidase(GSH-PX) were determined. RESULTS To the D-galactose-induced aging mice,Butyl-phthalide (20,40mg.kg-1,ig) reduced the content of LPO,LF, inhibited the decrease of body weight and the weight of thymus and spleen,enhance the activities of SOD in brain,CAT in erythrocyte and whole blood GSH-PX. CONCLUSION Butyl-phthalide shows a remarkable anti-aging effect on the D-galactose-induced aging mice.

2.1.2儀器與試劑：
2.1.2.1儀器：721分光光度計(jì)、離心機(jī)、恒溫水浴鍋、勻漿器
2.1.2.2試劑：SOD反應(yīng)液配置：
2.1.2.2.1 NBT：7.5×10-6M 配成10倍母液，取0.06133gNBT加少量PH7.8 50mM PBS稍加熱溶解（60-70℃）定容至100ml；
2.1.2.2.2 Met：13×10-3M 配成10倍母液，取1.9397gMet加約50mlPH7.8 50mM PBS，加熱溶解，最后定容至100ml；
2.1.2.2.3 EDTA：292.25×100×10-9M 配成100倍母液，0.29mg溶于100mlPH7.8 50mM PBS或者2.9mg溶于1000mlPH7.8 50mM PBS
